M6PR: The Trafficker of Lysosomal Enzymes
The Cation-Dependent Mannose-6-Phosphate Receptor (M6PR), also known as CD-M6PR, is a transmembrane receptor that shuttles from the trans-Golgi network (TGN) to endosomes. M6PR binds newly synthesized lysosomal enzymes that have been tagged with a mannose-6-phosphate (M6P) signal in the Golgi. M6PR binds the enzyme in the TGN and then traffics it via clathrin coated vesicles to endosomes. Once in the endosome, the enzyme dissociates from the receptor due to the low pH and the receptor is recycled back to the TGN for another round of trafficking. This M6PR-mediated trafficking is the major pathway for lysosomes to be populated with the correct hydrolases.

Background
This gene encodes a member of the P-type lectin family. P-type lectins play a critical role in lysosome function through the specific transport of mannose-6-phosphate-containing acid hydrolases from the Golgi complex to lysosomes. The encoded protein functions as a homodimer and requires divalent cations for ligand binding. Alternatively spliced transcript variants encoding multiple isoforms have been observed for this gene. A pseudogene of this gene is located on the long arm of chromosome X.
