The Human ADAM9 Intron 11 Primers provide a specific and optimized solution for amplifying DNA obtained through chromatin immunoprecipitation (ChIP). This product contains a pre-mixed set of forward and reverse PCR primers designed to target intron 11 of the human ADAM9 (ADAM Metallopeptidase Domain 9) gene. ADAM9 is a disintegrin and metalloproteinase involved in cell adhesion, migration, and proteolytic processing of various proteins, often implicated in cancer progression. Investigating intronic regions can provide insights into chromatin states or RNA polymerase progression within the gene body. These primers are optimized for use in SYBR® Green quantitative real-time PCR and are validated with Enzymatic Chromatin IP Kits and ChIP-validated antibodies.

  • The primary function of primers in chromatin immunoprecipitation (ChIP) experiments is to serve as specific probes for detecting and quantifying particular DNA fragments enriched after immunoprecipitation, thereby revealing protein-DNA interactions.
  • Workflow
  • 1. Label PCR tubes/plates for duplicates, no-DNA control, and a 2% total input chromatin DNA serial dilution.
    2. Add 2 μl ChIP DNA sample per well/tube.
    3. Prepare and add 18 μl master PCR reaction mix (Nuclease-free H2O, Primers, SYBR® Green).
    4. Run 40 cycles of PCR: 95°C for 3 min; then 95°C for 15 sec, primer-specific temp for 60 sec (repeated).
    5. Analyze results with real-time PCR software.

  • Introduction
  • This gene encodes a member of the ADAM (a disintegrin and metalloprotease domain) family. Members of this family are membrane-anchored proteins structurally related to snake venom disintegrins, and have been implicated in a variety of biological processes involving cell-cell and cell-matrix interactions, including fertilization, muscle development, and neurogenesis. The protein encoded by this gene interacts with SH3 domain-containing proteins, binds mitotic arrest deficient 2 beta protein, and is also involved in TPA-induced ectodomain shedding of membrane-anchored heparin-binding EGF-like growth factor. Several alternatively spliced transcript variants have been identified for this gene.
