The Human HoxA2 Promoter Primers are designed for amplifying DNA isolated through chromatin immunoprecipitation (ChIP). This product includes optimized forward and reverse PCR primers specific to the promoter region of the human HoxA2 gene. HoxA2 is a homeobox gene involved in orchestrating embryonic development, particularly in hindbrain patterning and skeletal formation. Its precise transcriptional regulation is critical for proper development. These primers are optimized for SYBR® Green quantitative real-time PCR and are thoroughly validated with Enzymatic Chromatin IP Kits and ChIP-validated antibodies. They enable detailed investigation of epigenetic control over developmental gene expression.

  • The primary role of primers in chromatin immunoprecipitation (ChIP) assays is to act as precise probes for identifying and quantifying particular DNA sequences that have been enriched through immunoprecipitation, thereby uncovering protein-DNA interactions.
  • Workflow
  • 1. Label PCR tubes/plates for duplicates, no-DNA control, and a 2% total input chromatin DNA serial dilution.
    2. Add 2 μl ChIP DNA sample per well/tube.
    3. Prepare and add 18 μl master PCR reaction mix (Nuclease-free H2O, Primers, SYBR® Green).
    4. Run 40 cycles of PCR: 95°C for 3 min; then 95°C for 15 sec, primer-specific temp for 60 sec (repeated).
    5. Analyze results with real-time PCR software.

  • In vertebrates, the genes encoding the class of transcription factors called homeobox genes are found in clusters named A, B, C, and D on four separate chromosomes. Expression of these proteins is spatially and temporally regulated during embryonic development. This gene is part of the A cluster on chromosome 7 and encodes a DNA-binding transcription factor which may regulate gene expression, morphogenesis, and differentiation. The encoded protein may be involved in the placement of hindbrain segments in the proper location along the anterior-posterior axis during development.
