The Human SF3B3 Exon 1 Primers offer a reliable tool for amplifying DNA isolated via chromatin immunoprecipitation (ChIP). This product features a pre-mixed set of forward and reverse PCR primers specifically targeting exon 1 of the human SF3B3 (Splicing Factor 3b Subunit 3) gene. SF3B3 is a core component of the U2 snRNP, essential for pre-mRNA splicing. - Fundamentally, primers in ChIP assays act as highly specific probes. After the initial immunoprecipitation step, where target proteins (and their bound DNA) are isolated, the challenge becomes identifying exactly which DNA sequences were pulled down. This is where primers shine. Designed to be complementary to very precise regions of DNA, they bind exclusively to the sequences of interest. This specificity is crucial, ensuring that only the genuinely enriched DNA fragments are amplified and detected.
- Workflow
- 1. Label PCR tubes/plates for duplicates, no-DNA control, and a 2% total input chromatin DNA serial dilution.
2. Add 2 μl ChIP DNA sample per well/tube.
3. Prepare and add 18 μl master PCR reaction mix (Nuclease-free H2O, Primers, SYBR® Green).
4. Run 40 cycles of PCR: 95°C for 3 min; then 95°C for 15 sec, primer-specific temp for 60 sec (repeated).
5. Analyze results with real-time PCR software.

- Introduction
- This gene encodes subunit 3 of the splicing factor 3b protein complex. Splicing factor 3b, together with splicing factor 3a and a 12S RNA unit, forms the U2 small nuclear ribonucleoproteins complex (U2 snRNP). The splicing factor 3b/3a complex binds pre-mRNA upstream of the intron's branch site in a sequence independent manner and may anchor the U2 snRNP to the pre-mRNA. Splicing factor 3b is also a component of the minor U12-type spliceosome. Subunit 3 has also been identified as a component of the STAGA (SPT3-TAF(II)31-GCN5L acetylase) transcription coactivator-HAT (histone acetyltransferase) complex, and the TFTC (TATA-binding-protein-free TAF(II)-containing complex). These complexes may function in chromatin modification, transcription, splicing, and DNA repair.
