The Human SUB1 Promoter Primers are designed for amplifying DNA isolated through chromatin immunoprecipitation (ChIP). This product includes optimized forward and reverse PCR primers specific to the promoter region of the human SUB1 (Suppressor of RNA Polymerase II Transcription 1) gene. SUB1 is a general transcription factor involved in both positive and negative regulation of RNA polymerase II transcription. Studying its promoter is crucial for understanding its transcriptional control and its role in gene expression. These primers are optimized for SYBR® Green quantitative real-time PCR and are thoroughly validated with Enzymatic Chromatin IP Kits and ChIP-validated antibodies.

  • Once bound to their target DNA, primers serve as the starting points for DNA amplification, typically through quantitative Polymerase Chain Reaction (qPCR) or next-generation sequencing (ChIP-seq). In qPCR, the primers enable the exponential copying of the target DNA, allowing for its precise quantification. The amount of amplified DNA is directly proportional to the initial amount of target DNA present in the immunoprecipitated sample. This quantitative measure is incredibly valuable, as it not only tells us if a protein binds to a specific DNA region, but also how strongly or how frequently that binding occurs under different conditions.
    For ChIP-seq, primers are used to generate libraries of DNA fragments that are then sequenced. This provides an even broader, genome-wide view of protein-DNA binding sites, and primers are essential for preparing these fragments for the sequencing process.
  • Workflow
  • 1. Label PCR tubes/plates for duplicates, no-DNA control, and a 2% total input chromatin DNA serial dilution.
    2. Add 2 μl ChIP DNA sample per well/tube.
    3. Prepare and add 18 μl master PCR reaction mix (Nuclease-free H2O, Primers, SYBR® Green).
    4. Run 40 cycles of PCR: 95°C for 3 min; then 95°C for 15 sec, primer-specific temp for 60 sec (repeated).
    5. Analyze results with real-time PCR software.
