ENO3: The Muscle-Specific Glycolytic Enzyme

Enolase 3 (ENO3), also known as beta-enolase or Eno-β, is the muscle-specific isoform of the glycolytic enzyme enolase, a key enzyme in the conversion of glucose to pyruvate. It catalyzes the conversion of 2-phosphoglycerate (2-PG) to phosphoenolpyruvate (PEP), a high-energy intermediate, leading to the production of ATP. ENO3 is highly expressed in fast and slow skeletal muscle fibers, where it plays a critical role in supplying the high energy demand required for muscle contraction. Genetic mutations in ENO3 can lead to a rare glycogen storage disease (GSD XIII) that manifests as exercise intolerance and muscle breakdown (rhabdomyolysis).

Background

This gene encodes one of the three enolase isoenzymes found in mammals. This isoenzyme is found in skeletal muscle cells in the adult where it may play a role in muscle development and regeneration. A switch from alpha enolase to beta enolase occurs in muscle tissue during development in rodents. Mutations in this gene have be associated glycogen storage disease. Alternatively spliced transcript variants encoding different isoforms have been described.
