S1PR1: A Promising Target for Immunomodulatory Therapy
A member of the G protein-coupled receptor (GPCR) family, S1PR1 (sphingosine-1-phosphate receptor 1) is essential in controlling important physiological processes, including immune cell motility. An important process driving immune surveillance is the transfer of lymphocytes from lymphoid organs to systemic circulation, controlled by this receptor. Apart from its immunological function, S1PR1 affects oncogenesis and cardiovascular pathology, maintenance of endothelial barrier integrity, and helps in vascular morphogenesis. The rise of this receptor as a therapeutic target in autoimmune diseases has been driven by its effect on lymphocyte trafficking.

Background
The protein encoded by this gene is structurally similar to G protein-coupled receptors and is highly expressed in endothelial cells. It binds the ligand sphingosine-1-phosphate with high affinity and high specificity, and suggested to be involved in the processes that regulate the differentiation of endothelial cells. Activation of this receptor induces cell-cell adhesion. Alternative splicing results in multiple transcript variants.
