TP53: The Guardian of the Genome
Tumor Protein p53 (TP53) is a transcription factor that functions as a convergence point in a complex network of cellular stress response pathways. In the absence of stress, p53 is kept at extremely low levels by its negative regulator MDM2, which ubiquitinates the protein and targets it for degradation. However, under conditions of cellular stress, such as DNA damage or oncogene activation, p53 is rapidly stabilized and activated. The stabilized p53 protein then moves to the nucleus, where it activates a transcriptional program that generally has one of three possible outcomes: a reversible cell cycle arrest to allow time for DNA repair, irreversible cell cycle arrest (senescence), or programmed cell death (apoptosis). Loss-of-function mutations in TP53 are the most common genetic alteration found in human cancer, present in over 50% of all human tumors.

Background
This gene encodes a tumor suppressor protein containing transcriptional activation, DNA binding, and oligomerization domains. The encoded protein responds to diverse cellular stresses to regulate expression of target genes, thereby inducing cell cycle arrest, apoptosis, senescence, DNA repair, or changes in metabolism. Mutations in this gene are associated with a variety of human cancers, including hereditary cancers such as Li-Fraumeni syndrome. Alternative splicing of this gene and the use of alternate promoters result in multiple transcript variants and isoforms. Additional isoforms have also been shown to result from the use of alternate translation initiation codons from identical transcript variants.
