This antibody-cytokine fusion protein was achieved by conjugating/fusing the Anti-EpCAM IgG (N to Q) to IL2. It was expressed in CHO and purified with affinity chromatography. The immunocytokine retains the ability to bind the EpCAM as well as the biological activity of IL2. HuKS (N to Q)-IL2 contains an amino acid substitution in the CH2 domain of the antibody Fc gamma 1 constant region that eliminates N-linked glycosylation. The amino acid sequence is changed from QYNSTYR to QYQSTYR, thereby additionally eliminating a potential T cell epitope. This immunocytokine was designed for treating solid tumor.
