This research investigates the molecular mechanisms underlying triple-negative breast cancer (TNBC) cells' ability to specifically metastasize to the lungs. Using differential/suppression subtractive chain (D/SSC) technique, the researchers identified a 24-gene signature induced by geminin overexpression (GemOE) that promotes lung-specific metastasis. Five key genes from this signature (LGR5, HAS2, CDH11, NCAM2, and DSC2) were associated with worsened lung metastasis-free survival in TNBC patients. The study demonstrates that LGR5-induced β-catenin signaling and stemness in TNBC cells are geminin-overexpression dependent. Additionally, the researchers discovered RSPO2 expression exclusively in mouse lung tissue and identified a novel extravasation receptor complex consisting of CDH11, CD44v6, c-Met, and AXL on GemOE/LGR5+ TNBC lung metastatic precursors.

These antibodies were critical for in vivo neutralizing experiments that demonstrated blocking any component of the extravasation receptor complex prevented geminin overexpressing/LGR5+ TNBC lung metastasis. This contribution helped establish the therapeutic potential of targeting geminin in combination with components of the extravasation receptor complex to reduce TNBC lung metastasis and improve patient outcomes.
