Western blotting analysis was followed by SDS 5% to 20% PAGE under nonreducing (NR) and reducing IR) conditions. Each lane represents 33 Fg of a whole tryptic digest of vWF (E/S ratio indicated below). C, control (undigested vWF).

An aliquot of each fragment was applied onto a nitrocellulose membrane in a circular area. After drying, the membrane was treated with a solution of the anti-vWF monoclonal antibody NMC-4 (5 mg/ml) followed by fluorographic detection of the bound antibody using an alkaline phosphatase-conjugated rabbit antimouse IgG. Note that the antibody reacts well with the cyclic fragments containing the intact Cys509–Cys695 disulfide bond (C) but only minimally after reduction and alkylation of Cys residues (R/A).

  • The VWF antibody has been reported in applications of Blocking, Inhibition, Western Blot, Dot Blot.
    For Inhibiton: Formalin-ked platelets were prepared as previously described and used at a concentration of 10⁸ platelets/mL in an incubation volume of 125 pL. [¹²⁵I]vWF or [¹²⁵I]AS-vWF was added at a concentration of 5 or 20 µg/mL in the presence of competing ligands or MoAbs at concentrations described in Results. For vWF binding, ristocetin or botrocetin was used at a final concentration of 1 mg/mL or 15 µg/mL. Binding was measured after 30 minutes of incubation at room temperature, and nonspecific binding was determined in the presence of a 50-fold excess of unlabeledvWF or AS-vWF. Specific binding was calculated by subtracting nonspecific binding from the total.
