This research investigates the role of MALAT1, a long non-coding RNA, in hepatocellular carcinoma (HCC) development and progression. The study demonstrates that MALAT1 is aberrantly overexpressed in HCC tissues and cell lines compared to normal liver tissues, with expression levels correlating with poor cellular differentiation and disease progression. Through shRNA-mediated silencing of MALAT1, the researchers showed that MALAT1 interacts directly with β-catenin and modulates the Wnt/β-catenin signaling pathway, thereby promoting cancer stemness characteristics. MALAT1 silencing reduced the CD133highCD90high HCC stem cell population, suppressed tumorsphere formation capacity, inhibited invasion and metastasis, and significantly reduced tumor growth in xenograft mouse models.
