The Anti-CD3E Antibody Prodrug, Protease Activated (VS-1025-YC3) is an antibody prodrug comprised of a recombinant hamster antibody targeting mouse CD3E, linked to a masking peptide that conceals the active antigen-binding site via a protease-cleavable linker. The mask peptide is recombinantly attached to the amino terminus of either the light or heavy chain of the antibody, designed to block access to the antigen-binding site and physically prevent the antibody from binding to CD3E protein. Once the masking peptide is cleaved by matrix metalloproteinases (MMPs), the prodrug is activated, allowing the unmasked monoclonal antibody component to bind to CD3E effectively. CD3 epsilon is a necessary component of the T-cell Receptor (TCR) complex, which is essential for the T-cell to recognize and eliminate tumor cells; therefore, its relevance is purely immunotherapeutic. In cancer therapy, the murine CD3 complex is strategically engaged by bispecific antibodies that simultaneously bind both a tumor antigen and CD3, forcibly activating the T-cell to execute a precise, targeted attack on the tumor.

Introduction
The protein encoded by this gene is the CD3-epsilon polypeptide, which together with CD3-gamma, -delta and -zeta, and the T-cell receptor alpha/beta and gamma/delta heterodimers, forms the T-cell receptor-CD3 complex. This complex plays an important role in coupling antigen recognition to several intracellular signal-transduction pathways. The genes encoding the epsilon, gamma and delta polypeptides are located in the same cluster on chromosome 11. The epsilon polypeptide plays an essential role in T-cell development. Defects in this gene cause immunodeficiency. This gene has also been linked to a susceptibility to type I diabetes in women.
