  Briefly, lymphoid cells were washed twice with cold BSS, centrifuged (1000g, 1 min), and approximately 1 X 10⁶ cells were incubated for 30 min on ice with saturating amounts of primary antibody in l-ml polystyrene microcentrifuge tubes (Analytical Lab Associates, NY). The cells were washed twice with ice-cold PBS-BSA-azide (0.01 M phosphate, 2.0% BSA, 0.05% sodium azide, pH 7.2). Twenty five microliters of 1: 10 fluorescein isothiocyanate (FITC)-conjugated affinity purified F(ab')₂ goat anti-mouse IgG (heavy and light chain specific) (FITC GaM) (Cappel, Malvern, PA) was added and samples were vortexed and incubated for 20 min at 4°C. Samples were washed twice with ice-cold PBS-BSA-azide and resuspended in 125 μL PBS-BSA-azide. Negative controls consisted of cells stained with mouse IgG (Sigma) and FITC GaM. Samples were analyzed on an EPICS 751 flow cytometer (Coulter Electronics, Hialeigh, FL), equipped with an argon ion laser running at 400 mW and 488 nm. Cell populations of interest were electronically gated based on light scatter characteristics, both forward angle and 90° angle. Analysis was based on 10,000 cells using the Integra program on an Easy 2 data analysis system (Coulter Electronics).
