The Human Timeless Intron 1 Primers offer a reliable tool for amplifying DNA isolated via chromatin immunoprecipitation (ChIP). This product features a pre-mixed set of forward and reverse PCR primers specifically targeting intron 1 of the human Timeless (TIM) gene. Timeless is a crucial component of the circadian clock machinery and is also involved in DNA damage response. Investigating intronic regions can provide insights into chromatin states within the gene body or transcriptional elongation. These primers are optimized for use in SYBR® Green quantitative real-time PCR and are validated with Enzymatic Chromatin IP Kits and ChIP-validated antibodies.

- Primers in ChIP assays are highly specific probes. Their main function is to detect and quantify particular DNA sequences that have been isolated via immunoprecipitation, ultimately revealing crucial protein-DNA interactions.
- Workflow
- 1. Label PCR tubes/plates for duplicates, no-DNA control, and a 2% total input chromatin DNA serial dilution.
2. Add 2 μl ChIP DNA sample per well/tube.
3. Prepare and add 18 μl master PCR reaction mix (Nuclease-free H2O, Primers, SYBR® Green).
4. Run 40 cycles of PCR: 95°C for 3 min; then 95°C for 15 sec, primer-specific temp for 60 sec (repeated).
5. Analyze results with real-time PCR software.

- The protein encoded by this gene is highly conserved and is involved in cell survival after damage or stress, increase in DNA polymerase epsilon activity, maintenance of telomere length, and epithelial cell morphogenesis. The encoded protein also plays a role in the circadian rhythm autoregulatory loop, interacting with the PERIOD genes (PER1, PER2, and PER3) and others to downregulate activation of PER1 by CLOCK/ARNTL. Changes in this gene or its expression may promote prostate cancer, lung cancer, breast cancer, and mental disorders.
