SLC7A11: The Key Transporter for Cystine Uptake
SLC7A11 is a multi-pass transmembrane protein that serves as the specific exchange subunit for the system, facilitating the extracellular uptake of cystine in exchange for intracellular glutamate. Once transported into the cell, cystine is rapidly reduced to cysteine, the rate-limiting precursor for intracellular glutathione (GSH) synthesis, which is the major cellular antioxidant. Cancer cells rely on SLC7A11 to maintain high intracellular GSH levels for protection from oxidative stress and chemotherapeutics. Inhibition of SLC7A11 can trigger ferroptosis, making it a prime therapeutic target.

Background
This gene encodes a member of a heteromeric, sodium-independent, anionic amino acid transport system that is highly specific for cysteine and glutamate. In this system, designated Xc(-), the anionic form of cysteine is transported in exchange for glutamate. This protein has been identified as the predominant mediator of Kaposi sarcoma-associated herpesvirus fusion and entry permissiveness into cells. Also, increased expression of this gene in primary gliomas (compared to normal brain tissue) was associated with increased glutamate secretion via the XCT channels, resulting in neuronal cell death.
