The TNF-FuP (fusion protein) provides only a weak stimulus for lymphocyte proliferation and has no effect on LAK cells. However, it supports the activation of macrophages (Mfs) and, to a minor extent, cytotoxic T lymphocytes (CTLs).

What is the design purpose of this immunocytokine?

This immunocytokine was designed for research into treating CD30-positive lymphoma. It conjugates Anti-CD3 IgG to TNF, retaining the ability to bind CD3 while delivering the biological activity of Tumor Necrosis Factor.

Anti-CD3 Antibody-Cytokine Fusion Protein, TNF-IgG

This antibody-cytokine fusion protein was achieved by conjugating/fusing the Anti-CD3 IgG to TNF. It was expressed in CHO and purified with affinity chromatography. The immunocytokine retains the ability to bind the CD3 as well as the biological activity of TNF. TNF-FuP provided a weak stimulus for lymphocyte proliferation, but it had no effect on LAK cells. However, it supported activation of Mfs and, to a minor extent, of CTLs. This immunocytokine was designed for treating CD30-positive lymphoma.

Cytokines 1 Description
Tumor necrosis factor (TNF), formerly known as TNFα or TNF alpha, is the best-known member of this class. TNF is a monocyte-derived cytotoxin that has been implicated in tumor regression, septic shock, and cachexia. The protein is synthesized as a prohormone with an unusually long and atypical signal sequence, which is absent from the mature secreted cytokine. A short hydrophobic stretch of amino acids serves to anchor the prohormone in lipid bilayers. Both the mature protein and a partially processed form of the hormone can be secreted after cleavage of the propeptide.

Macrophage Activation Support

While lymphocyte proliferation was weak as expected, this TNF-IgG fusion strongly supported the activation of macrophages in our assays. It has been useful for dissecting the roles of different cell types in CD30-positive lymphoma research.

Targets CD3 Effectively

This product binds CD3 efficiently and delivers the TNF payload as designed. We have used it to investigate the tumoricidal effects of TNF in a targeted manner, specifically looking at regression in solid tumor models.
