Twenty microliters of CWPs (about 50 to 100 ng of total protein) were loaded on a 12% SDS-PAGE gel, separated and blotted onto nitrocellulose membrane. After blocking with 5% (w/v) skimmed milk, the membrane was incubated with 50 nmol/L bacterially expressed FvCA4 followed by the addition of a mouse anti-His antibody and a AP-conjugated goat anti-mouse IgG antibody. The colorimetric reaction was carried out by adding BCIP/NBT substrate. M, protein molecular weight standards. Lanes 1 to 4, the CWPs of Fusarium verticillioides, Fusarium asiaticum, Aspergillus flavus, and Sclerotinia sclerotiorum.
