ACVR1: A Potential Target in Cancer Therapy

ACVR1 is a receptor that plays a key role in bone morphogenetic protein signaling. It is linked to fibrodysplasia ossificans progressiva (FOP), a rare disorder of abnormal bone formation. ACVR1 mutations are associated with cardiac and reproductive system abnormalities. Recently, ACVR1 has been implicated as a cancer driver gene in a highly aggressive pediatric brain tumor DIPG. This shows great potential to target ACVR1 to develop therapeutics for cancer therapy.

Background

Activins are dimeric growth and differentiation factors that belong to the transforming growth factor-beta (TGF-beta) superfamily of structurally related signaling proteins. Activins signal through a heteromeric complex of receptor serine kinases which include at least two type I (I and IB) and two type II (II and IIB) receptors. These receptors are all transmembrane proteins, composed of a ligand-binding extracellular domain with a cysteine-rich region, a transmembrane domain, and a cytoplasmic domain with predicted serine/threonine specificity. Type I receptors are essential for signaling, and type II receptors are required for binding ligands and expression of type I receptors. Type I and II receptors form a stable complex after ligand binding, resulting in the phosphorylation of type I receptors by type II receptors. This gene encodes activin A type I receptor which signals a particular transcriptional response in concert with activin type II receptors. Mutations in this gene are associated with progressive fibrodysplasia ossificans.

Human Anti-ACVR1 Recombinant Antibody (HPAB-0707LY) (CAT#: HPAB-0707LY)
I recently purchased the anti-ACVR1 antibody from Creative Biolabs for use in our lab's WB experiments, and I am thoroughly impressed with its performance. Firstly, the shipping and packaging were prompt and top-notch. The datasheet provided was detailed and included comprehensive information on recommended dilutions, storage conditions, and application specifics. We used the anti-ACVR1 antibody at the suggested concentration for our WB protocol to detect the expression of ACVR1 in lysates from both human and mouse cell lines. Surprisingly, the anti-ACVR1 antibody provided clear, distinct bands at the expected molecular weight with minimal background noise. The results were outstanding.
