SSEA-4 is a ganglioside which consists of a glycosphingolipid (GSL) containing a terminal sialic acid residue (N-acetylneuraminic acid). It is synthesized from SSEA-3 by the enzyme ST3 beta-galactoside alpha-2,3-sialyltransferase 2 (ST3GAL2). The expression of SSEA-4 undergoes qualitative and quantitative changes during development, differentiation and tumorigenesis. During human preimplantation development, SSEA-3 and -4 are first observed on the pluripotent cells of the inner cell mass and are lost upon differentiation. In addition, human germ stem cells in the testis and ovary express SSEA-4.
