TLR9 is an endosomal receptor expressed on B cells, macrophages, and dendritic cells. It recognizes unmethylated CpG motifs in bacterial and viral DNA. When TLR9 binds to CpG - containing DNA, it recruits the MyD88 adaptor protein. This initiates a signaling cascade that activates NF - κB and other transcription factors, leading to the production of cytokines such as interleukin - 6 (IL - 6), interleukin - 12 (IL - 12), and type I interferons. TLR9 - mediated immune responses are important for the defense against intracellular pathogens and also play a role in the activation of the adaptive immune system.
