Recombinant Mouse Antibody (JEL42) is capable of binding to E. coli ptsH, expressed in Chinese Hamster Ovary cells (CHO).

Antibodies were serially diluted into the microtiter plates and incubated, and then the unbound antibody was washed off. Bound antibody was determined by using 125I-labeled sheep anti-mouse IgG antibody. The monoclonal antibodies were Jel42 from culture fluid (*), Jel44 from culture fluid (A), and Jel42 purified
from ascites fluid (A).

The results presented are the inhibition of antibody binding to the HPr bound to microtiter plates as a function of the amount of competing protein added to the
wells. The antibody preparation used in this experiment was a 1:4 dilution of the Jel42 from culture fluid. Samples: HPr (○); extracts of S. typhimurium SB3507 grown on nutrient broth (Δ), nutrient broth with glucose (A), or nutrient broth with fructose -- -, 0); extracts of S. typhimurium SB2226 grown on nutrient broth (O) or nutrient broth with fructose (O).

Competition SPRIA assays were carried out that the antibody preparation for Jel42 was from ascites fluid (1:400). Wild-type HPr from E. coli (solid symbols) and HPr from S. typhimurium SB3093 (open symbols) were tested with Jel42 (triangles) and Jel44 (circles).
