Anti-HER2/neu Immunocytokine, scFv (4D5)-IL15 (ACFP-SH011)
This antibody-cytokine fusion protein was achieved by conjugating/fusing the Anti-HER2/neu scFv to IL15. It was expressed in CHO and purified with affinity chromatography. The immunocytokine retains the ability to bind the HER2/neu as well as the biological activity of IL15. The goal of this antibody-cytokine fusion protein to cancer therapy is to concentrate the cytokine in the tumor microenvironment and in so doing directly enhance the tumoricidal effect of the antibody and/or enhance the host immune response (T-cell, B-cell or NK) against the tumor. This immunocytokine was designed for treating HER-2/neu over-expressing cancer.

- Interleukin 15 (IL-15) is a cytokine with structural similarity to IL-2. Like IL-2, IL-15 binds to and signals through a complex composed of IL-2/IL-15 receptor beta chain (CD122) and the common gamma chain (gamma-C, CD132). IL-15 is secreted by mononuclear phagocytes (and some other cells) following infection by virus (es). This cytokine induces cell proliferation of natural killer cells; cells of the innate immune system whose principal role is to kill virally infected cells.
