The anti-ERBB2 pH-dependent antibody (VS-0825-YC117) targets human ErbB2 in a pH-dependent manner that affinity is kept high at neutral pH but becomes weaker when pH is lowered, such as in the acidic environment of the endosomes. The anti-ERBB2 acid-switched antibody was engineered by incorporating histidine residues into their variable regions. The introduction of histidine substitutions into its CDRs allowed pH-responsive antigen-binding leading to enhanced ERBB2 clearance. The antibody works in cycles of ERBB2 binding in the extracellular fluid and ERBB2 releasing in acidified endosomes. Human ErbB2, also known as HER2, is a receptor tyrosine kinase that belongs to the EGFR family. Its overexpression is a key driver of certain aggressive breast and ovarian cancers, making it a major therapeutic target.
