  • This antibody has been reported for use in Western Blot, Immunoprecipitation and Flow Cytometry.
    FC: for flow-cytometry analyses, aliquots of 1×10⁶ PBLs or sorted populations were incubated with 50 ml mAb supernatant for 30 min at 4˚C, washed, and incubated with 50 ml goat anti-mouse secondary Abs conjugated to PE diluted 1:80 (v/v) in cfRPMI or FITC diluted 1:40 (v/v) in cfRPMI for 30 min at 4˚C. After washing, cells were suspended in 0.3 ml cfRPMI and analyzed for single or two-color staining by flow cytometry.
    IP: Immunoprecipitations were performed using Sepharose beads that were prepared in a stepwise fashion. First, 1 g lyophilized cyanogen bromide (CNBr)-activated Sepharose 4B beads was washed in 200 ml 1 mM HCl and resuspended in 5 ml coupling buffer containing 0.5 mg goat anti-mouse IgM for 1 h. After washing in coupling buffer, any remaining active groups were blocked by incubating the coupled beads in 0.1 M Tris-HCl (pH 8) for 2 h, and the beads were washed in alternating high- and low-pH buffers to remove excess uncoupled ligand. Second, 8 ml anti-catfish Igd mAb 7D11 hybridoma supernatant, diluted 1:1 in low-stringency buffer (100 mM NaCl, 20 mM Tris-HCl [pH 7.7]), was added to the goat antimouse IgM-coupled beads, which were allowed to incubate with end-toend rotation overnight at 4˚C. After incubation, the beads were washed in low-stringency buffer (total of 30 ml added in six aliquots) and resuspended in 4 ml low-stringency buffer.
