Mouse Anti-IgL Recombinant Antibody (clone 3F12) (CAT#: FAMAB-0034-CN)

The monoclonal antibody 3F12 reacts with catfish IgL F isotype. This antibody has specificities for the catfish equivalents of kappa and lambda chains.

PBL were isolated from a G14D-CCV immunized fish 3 d after a booster immunization. An aliquot of these cells was analyzed for IgL chain staining with anti-IgL F (3F12) and IgL G (11A2) mAbs, followed by goat anti-mouse IgG1 FITC and goat anti-mouse IgG2b RPE. The remainder of the cells were sequentially magnetically sorted with 3F12 and 11A2 mAbs and separated into IgL F−/IgL G−, IgL F+ or IgL G+, and IgL F+ and IgL G+ cell populations. Each cell population was then stained with CC41 mAb directly conjugated to Alexa Fluor 488. Percentages of positive cells are indicated in the upper right corner, and staining with the isotype control Ab 1.14 is outlined in gray.

  • This antibody has been reported for use in Immunofluorescence and Flow Cytometry.
    FC: PBL were isolated from G14D and G14D-CCV–immunized fish at the indicated time points, resuspended at 1×10⁷ cells/ml in cfRPMI, and analyzed for CC41 staining using flow cytometry analysis. For single-color flow cytometry, 1×10⁶ cells were incubated with 50 ml mAb supernatant for 30 min on ice, washed with cfRPMI, and incubated with 50 ml goat anti-mouse IgG1-PE (1:80 v/v in cfRPMI) for 30 min on ice. After washing, samples were resuspended in 500 ml cfRPMI and analyzed using the FACScan. The 3F12⁺ /11A2⁺, 3F12⁺ /11A22⁺ 3F122/11A2⁺, and 3F122/11A22 cell populations were analyzed as above.
