The anti-Integrin αIIbβ3 pH-dependent antibody (clone 2F2) targets human Integrin αIIbβ3 in a pH-dependent manner, with its affinity remaining high at neutral pH but weakening at a lower pH, such as within the acidic environment of endosomes. The anti-Integrin αIIbβ3 acid-switched antibody was engineered by incorporating histidine residues into its variable regions. The introduction of histidine substitutions within its Complementarity-Determining Regions (CDRs) enables pH-responsive antigen binding, which leads to enhanced Integrin αIIbβ3 clearance. The antibody operates in a cyclical manner, binding Integrin αIIbβ3 in the extracellular fluid and releasing it in acidified endosomes. Integrin αIIbβ3, or glycoprotein IIb/IIIa, is a cell surface receptor exclusively expressed on platelets. It is essential for platelet aggregation and hemostasis, and is a key target for anti-platelet medications used to prevent blood clots.
