Human Anti-TSST-1 Recombinant Antibody (clone MS473) (CAT#: VS-0323-CJ62)

A total of 20 phage clones with potential binding to TSST-1 protein were identified, VS-0323-CJ62 had the one of the highest binding signals.

The wells coated with TSST-1 or bovine serum albumin (BSA) (as control) were individually incubated with MS473 or SP220, followed by mouse anti-human scFv polyclonal antibody (MPAb) or unimmunized mouse sera (UnMsera; control). Empty wells were used as the control. Next, the wells were incubated with goat anti-mouse IgG-horseradish peroxidase (HRP)-conjugated antibody (GtAb), followed by the addition of the TMB solution.

VS-0323-CJ62 exhibited a significant inhibitory effect (the IC50 value: 1.5 µM) on the proliferation of PBMCs and decreased the number of colony-forming cells.

The MS473 scFv reacted significantly with the peptides T1 and T2, which are located in the region of TSST-1 that interacts with the human TCR Vβ-chain. There was no significant reaction between the MS473 scFv and peptides M1 and M2, both of which are located within the region of TSST-1 that interacts with the MHC class II.
