Background

This gene encodes a member of the family of bifunctional 6-phosphofructo-2-kinase:fructose-2,6-biphosphatase enzymes. The enzyme forms a homodimer that catalyzes both the synthesis and degradation of fructose-2,6-biphosphate using independent catalytic domains. Fructose-2,6-biphosphate is an activator of the glycolysis pathway and an inhibitor of the gluconeogenesis pathway. Consequently, regulating fructose-2,6-biphosphate levels through the activity of this enzyme is thought to regulate glucose homeostasis. Multiple alternatively spliced transcript variants have been found for this gene.

The Rabbit Anti-PFKFB1 Recombinant Antibody worked well in our lab for Western blotting using HEK293 and HeLa cell lysates, detecting a clean band at the expected molecular weight. It also performed reliably in immunofluorescence, staining endogenous PFKFB1 in fixed A549 cells with minimal background. While we haven’t tested it in ELISA yet, the datasheet suggests it’s suitable for detection in non-denaturing assays. The antibody showed good lot-to-lot consistency, though optimization was needed for some tissue homogenates. A solid choice for basic research applications.

Rabbit Anti-PFKFB1 Recombinant Antibody (clone JU53-31) (CAT#: VS3-WK672)
This antibody performed reliably in our WB experiments, detecting clear bands in both mouse liver tissue lysates and cultured HepG2 cells. The signal was specific at the predicted size with minimal nonspecific binding after optimizing blocking conditions. While it worked well for WB, we noticed slightly weaker reactivity in heart tissue samples compared to liver. Overall, it's a dependable option for detecting PFKFB1 in common cell lines and tissues when proper controls are included.
