This antibody-cytokine fusion protein was achieved by conjugating/fusing the Anti-HER2/neu IgG to GM-CSF. It was expressed in CHO and purified with affinity chromatography. The immunocytokine retains the ability to bind the HER2/neu as well as the biological activity of GM-CSF. In addition, the anti-HER2/neu IgG3-(GM-CSF) protein was capable of activating J774.2 murine macrophage cells enhancing their direct cytotoxic activity and also enhancing antibody-dependent macrophage-mediated tumor cell lysis. The anti-HER2/neu IgG3-(GM-CSF) fusion protein will be useful in the treatment of human HER2/neu-expressing tumors. This immunocytokine was designed for treating HER-2/neu over-expressing cancer.
