Function Analysis

As a leading custom service provider in antibody field, Creative Biolabs provides comprehensive analysis services for complete function evaluation of your bispecific antibodies (BsAbs). Taking advantage of state-of-the-art instruments, our experienced staffs in analytical project design, equipment operation and data analysis can assist every customer to solve BsAb function analysis problems. BsAb is a novel kind of antibody format which has dual specificities targeting two different antigens simultaneously. Therefore, it is of great importance to assess their binding activity to antigens to ensure they have the desired function. For the BsAb with Fc fragment, it is strongly recommended to analysis the antibody dependent cellular cytotoxicity (ADCC) and Antibody-FcRn binding activity. Creative Biolabs offers BsAb function assay platforms tailored to meet every client’s requirements.

The function analysis of bispecific antibody we can offer includes:

  • Target binding affinity measurement
    • Assessment of two different antigens binding to BsAb independently
    • Assessment of two different antigens binding to BsAb simultaneously
  • ADCC assay
  • Antibody-FcRn binding analysis

Function Analysis

Our BsAb function assay platforms include, but not limited to:

  • SPR technology
Surface Plasmon Resonance (SPR) is a useful technique to measure the kinetics and binding affinities of biomolecular interaction in a label free, real-time monitoring manner. SPR detects changes in the refractive index in the immediate vicinity of the surface layer of a sensor chip.
  • ELISA-based assay
The enzyme-linked immunosorbent assay (ELISA) is a widely used plate-based assay technique for detecting and quantifying substances such as peptides, proteins, antibodies and hormones. ELISA-based assay is quite useful to verify antibody binding affinity to target antigen, by detecting the concentration of combined antibodies in antigen-antibody interaction system once the system reaching a balance stage.
  • Flow cytometry assay
Flow cytomertry approach can be utilized as a cell-based assay for antibody affinity measurement, which is especially helpful when the antibody targets a cell-surface antigen since this approach can ensure the proper antigen folding and display by employing whole cell in assay. The significant advantage of flow cytometry is that the antibody binding study can be performed without the purifing antigen or even knowing the antigen identity.
  • The 51Cr approach
The 51Cr approach is a frequently-used method to analyze the ADCC of IgG-like antibodies. Prior to performing the assay, isotope is pre-loaded into target cells. Once the 51Cr loaded target cells are lysed via ADCC process, 51Cr will be released into the supernatant. So measuring the radioactivity in the supernatant can indicate the extent of cell lysis.
