Heterogeneity in the N-glycans on antibodies and other biopharmaceuticals often result in poor process reproducibility and variable therapeutic efficacy. Non-glycosylated antibodies can be generated by producing in E. coli or mutating Asn297 to either Ala or Gln. N-glycans have important structural functions. They stabilize the CH2 domain and Non-glycosylated mAbs have been shown to be less thermally stable and more susceptible to proteolysis and unfolding. However, non-glycosylated mAbs have reduced effector functions while retain the FcRn interactions and long serum half-life of wild-type IgGs, which makes them appropriate for non-cancer targets to reduce potential adverse events. Several non-glycosylated IgGs or Fc platforms have been approved (e.g. Nplate®) and some candidates are in clinical trials. Besides, our scientists can shorten the N-glycosylation pathway in the Golgi by knocking-out and knocking-in specific glycosyltransferases or glycosidases to produce simplified-glycosylated antibodies. The simplified-glycosylated antibodies share similar folding and stability profiles meanwhile possess low initial clearance from serum and reduced effector functions relative to the wild-type antibodies.
