Tandem scFv-Fc Bispecific Antibody (BsAb)

Tandem scFv-Fc BsAb is an IgG-like antibody with two single chain variable domains targeting distinct antigens. When constructing a Tandem scFv-Fc BsAb expression vector, genes coding each single-chain variable domain are tandem arrayed to a human Fc gene. Hence, the heterodimer that consists of these two chains is the desired bispecific antibody.

Through carefully designed purification strategy, heterodimer can be successfully seperated from homodimers. However, there are also other approaches for a more efficient production. Creative Biolabs introduces "Knobs into Holes" approach into Fc-containing BsAb production. By substituting single amino acid in the opposing CH3 domains of each chain, the efficiency of heavy chain heterodimerization process is significantly improved. Furthermore, two or more Knob and Hole pairs can be introduced according to customer's requirements.
