The anti-CD22 pH-dependent antibody (clone Inotuzumab) is a recombinant monoclonal antibody that exhibits pH-dependent antigen binding to human CD22. By introducing histidine mutations that act as a pH-switch, the antibody demonstrates a preferential affinity for CD22 under acidic conditions compared to physiological pH. Using a yeast surface display system with pH-dependent screening pressure, this mAb is identified which shows a significant increase in dissociation from CD22 at pH 6.0. This pH-sensitive antibody retains its binding affinity for CD22 on tumor cells while exhibiting negligible binding to the receptor on normal cells at physiological pH. Human CD22 is a B-cell-specific co-receptor that acts as an inhibitory molecule to regulate B-cell signaling. It helps to maintain B-cell homeostasis and prevent an overly active immune response.
