This immunotoxin was achieved by conjugating/fusing the Anti-CD22 monoclonal antibody to ricin toxin. It was expressed in CHO and purified with affinity chromatography. The immunotoxin selectively binds to tumor cells. Then the ricin A chain is incorporated into the cell by receptor-mediated endocytosis, blocking ribosomal protein synthesis and ultimately leading to cell death. It was designed for treatment of B cell lymphoma.

Toxin Description
Ricin consist of two polypeptide chains, A and B, linked by means of disulfide bridge(s). Cytotoxicity is associated with the A chain and its 15 inhibition of protein synthesis in nucleated cells. The B chain is essentially a delivery vehicle for the A chain. The B chain recognizes polysaccharide units at the surface of cells and creates a high affinity interaction with such units. Once the B chain binds with polysaccharide units at 20" the celL surface, the A chain is incorporated into the cell, blocking ribosomal protein synthesis and ultimately leading tocell death. The use of ricin A chain is preferred in this invention.

Applications
This product is suitable for use in cytotoxicity assay, functional assay and immunotherapy.
