GYS1, or Glycogen Synthase 1, is a key enzyme involved in glycogen synthesis, particularly in muscle cells. It plays a crucial role in energy homeostasis by converting glucose into glycogen, which serves as an energy storage molecule. While GYS1 is essential for normal cellular function, it has also emerged as a potential therapeutic target for several conditions, such as glycogen storage diseases (GSDs) and neurodegenerative diseases. Furthermore, numerous investigations indicate that GYS1 might lead to tumor development and progression. Therefore, targeting GYS1 could be a novel therapeutic strategy for certain types of cancer or diseases.

The protein encoded by this gene catalyzes the addition of glucose monomers to the growing glycogen molecule through the formation of alpha-1,4-glycoside linkages. Mutations in this gene are associated with muscle glycogen storage disease. Alternatively, spliced transcript variants encoding different isoforms have been found for this gene.
