The anti-CD20 pH-dependent antibody (clone 18B12) is a recombinant monoclonal antibody that exhibits pH-dependent antigen binding to mouse CD20. By introducing histidine mutations that act as a pH-switch, the antibody demonstrates a preferential affinity for CD20 under acidic conditions compared to physiological pH. Using a yeast surface display system with pH-dependent screening pressure, this mAb is identified which shows a significant increase in dissociation from CD20 at pH 6.0. This pH-sensitive antibody retains its binding affinity for CD20 on tumor cells while exhibiting negligible binding to the receptor on normal cells at physiological pH. Mouse CD20 is a transmembrane protein expressed on B-cells and is the mouse equivalent of human CD20 (MS4A1). It shares a similar function in B-cell development and is a target in preclinical models of B-cell-related diseases.
