Although the B. subtilis manifests so many advantages as a biopharmaceutical producing host, the conventional standard B. subtilis system still suffers from the expensive inducer cost by using the positively regulated manP promoter of the mannose operon. In addition, other conventional induction systems also suffer a similar inducer drawback, such as the sugars, isopropyl-β-D-thiogalactopyranoside (IPTG), temperature shifts, acid shock, or ethanol, etc.

To addressing this critical technique bottleneck, the creative researchers were inspired by the existence of a few natural auto-inducible B. subtilis systems (the pst operon system, the gsiB system, and the lysine riboswitch system). In addition to the clear mechanisms of sugar activation and repression in the metabolism of B. subtilis, these pioneers created an incredible auto-inducible mechanism to fulfill the instantly growing demand for heterologous pharmaceuticals. Particularly, the auto-inducible system manipulates the mannose operon by deletion of the manP gene, which leads to a constitutive activation of the mannose operon without mannose. For instance, the TQ356 stands for a common strain of the auto-inducible system with an improved yield. Moreover, the high-cell-density culture technique furtherly improved the expression and accumulation efficiency.
