Anti-Human CD4 Recombinant Antibody (M-T412) (CAT#: TAB-166LC)

The antibody was generated using spleen cells from mice that were immunized with P815 mastocytoma cells transfected with human CD4 cDNA. The mice were further boosted with recombinant CD4. The antibody exhibited a strong affinity for CD4 receptor-bearing CEM cells and displayed a binding specificity similar to that of a murine anti-CD4 monoclonal antibody called MT151.

After 7 days, culture supernatant was harvested and assayed for human IgM by solid phase ELISA. Percent inhibition of IgM production was based on the concentration of IgM in the supernatant of PWM-stimulated cells incubated without CD4 mAb. Isotypematched control antibodies did not inhibit IgM production.

Plates were incubated for 3 days and culture supernatant was assayed for slL-2R by ELISA. Percent inhibition of slL-2R production was calculated by comparison with production in the absence of any mAb. Isotype-matched control antibodies, murine lgG2a (R11D10), chimeric lgG1 or lgG4 (c7E3), produced no inhibition. The average level of slL-2R produced by stimulated cells was 80 ng/10⁶ cells.

Cells were stained with FITC-labeled rabbit anti-mouse or anti-human IgG to detect the CD4 antigen and co-expression of the CD3 antigen was measured by staining with biotinyiated anti-Leu4, followed by streptavidin-PE. At each time point, the percent of cells staining positive for FITC or PE, or for both fluorophores, was determined by flow cytometry on a FACScan (Becton-Dickinson). The murine CD4 mAb, M-T413, served as a positive control.
