We reasoned that YADS2 and YADS3 recognized VEGF through a very similar mechanism, as they exhibited high sequence homology in their CDRs and bound to both human and murine VEGF. In contrast, YADS1 likely represented a unique mode of antigen recognition as it contained very different CDR sequences and did not recognize mVEGF.

In each panel, the rates of heat release upon binding are in the top plot and the integrated heats plotted against the molar ratio of Fab are in the bottom plot. Linear regression analysis yielded a molar ratio of 0.94G0.01 or 0.99G0.01 for Fab-YADS1 or Fab-YADS2 binding to hVEGF, respectively.

- This YADS1 antibody can be used in applications of WB, ELISA, Neut, FuncS, Thermodynamic characterization, SPR. It's recommended that the optimal antibody concentration, dilution, incubition time etc. are best to be carefully titrated in specific assays.
Thermodynamic characterization: KD values of Fab-YADS1 measured by ITC at 25°C (3 nM).
SPR: Serial dilutions of Fab proteins (3-500 nM) were injected, and binding responses on the hVEGF or mVEGF flow cell were corrected by subtraction of responses on a blank flow cell.
