The anti-MS4A1 pH-dependent antibody (VS-0825-YC17) is a recombinant monoclonal antibody that exhibits pH-dependent antigen binding to human MS4A1. By introducing histidine mutations that act as a pH-switch, the antibody demonstrates a preferential affinity for MS4A1 under acidic conditions compared to physiological pH. Using a yeast surface display system with pH-dependent screening pressure, this mAb is identified which shows a significant increase in dissociation from MS4A1 at pH 6.0. This pH-sensitive antibody retains its binding affinity for MS4A1 on tumor cells while exhibiting negligible binding to the receptor on normal cells at physiological pH. Human MS4A1, also known as CD20, is a transmembrane protein expressed on the surface of B-cells. It plays a crucial role in B-cell development and activation, making it a prominent target for antibody-based therapies in B-cell lymphomas and autoimmune diseases.
