This is a chimeric mouse-human monoclonal antibody (cMAb) against Burkholderia pseudomallei (BP). The cMAb CK3 reacts with lipopolysaccharides (38∼52 kDa in B. pseudomallei; 38∼60 kDa in B. thailandensis). This chimeric antibody was produced from hybridoma clone BP1 7F7.

For ELISA, the pIRES-H(L)-DHFR vectors containing chimeric H-chain (H1, H2, and H3) or L-chain (L1, L2, and L3) were cotransfected into CHO-DG44 cells using a LF-2000 reagent and the master seed cells were incubated for 3 days to get sufficient amounts of cMAbs before the DHFR-amplification process.

For Western blot analysis, 3 cMAb culture supernatants (cMAbs CK1, CK2, and CK3) were used at 1:5 dilutions.
