Anti-SDC1 Antibody-Cytokine Fusion Protein, IgG (B-B4)-IFNα2YNS (CAT#: VS-0624-FY79)
B-B4 represents a human monoclonal antibody that was designed to target the extracellular segment of CD138. The induced IFNα2 is released by infected cells and exerts its effects both locally and systemically on cells that possess particular cell surface receptors with the ability to bind to type I IFNs. The YNS mutations (H57Y, E58N, and Q61S) insert into a conserved IFNAR1 binding site that is in direct proximity to more variable residues of a putative tunable anchor. This product is an anti-human CD138-IFNα2YNS fusion protein, containing the V region of the anti-CD138 mouse antibody B-B4 and the human IgG1, κappa constant region. This IFNα2YNS mutant contains mutations at 3 positions, H57Y/E58N/Q61S. Human IFNα2YNS is fused to the C-terminus of the antibody via a SerGly4Ser peptide linker. The reference shows that the Anti-CD138-IFNα2YNS has greater activity than anti-CD138-IFNα2.

- IFNα2 is a protein secreted by cells infected with viruses, where it acts on other cells to inhibit viral infection. There are 3 types of interferons: interferon type I, interferon type II, and interferon type III. Type II interferons, also known as IFNγ, are produced by specific cells of the immune system. Unlike type I and type III interferons, IFNγ plays only a modest role in directly limiting viral infection. Type I and type III interferons act similarly. However, the effects of type III interferons (also known as IFNλ) are limited to epithelial cells, whereas type I interferons act on all cells of the body.

- The protein encoded by this gene is a transmembrane (type I) heparan sulfate proteoglycan and is a member of the syndecan proteoglycan family. The syndecans mediate cell binding, cell signaling, and cytoskeletal organization and syndecan receptors are required for internalization of the HIV-1 tat protein. The syndecan-1 protein functions as an integral membrane protein and participates in cell proliferation, cell migration and cell-matrix interactions via its receptor for extracellular matrix proteins. Altered syndecan-1 expression has been detected in several different tumor types. While several transcript variants may exist for this gene, the full-length natures of only two have been described to date. These two represent the major variants of this gene and encode the same protein.
