CD274 (cluster cluster 274), also known as PD-L1 (programmed death ligand 1) or B7-H1 (B7 homolog 1), is a protein encoded by the human CD274 gene in humans. CD274 is a 40 kDa type 1 transmembrane protein that is presumed to play a major role in suppressing the immune system during specific events such as pregnancy, tissue allografts, autoimmune diseases and other disease states such as hepatitis. Normally, the immune system responds to foreign antigens associated with exogenous or endogenous danger signals, triggering the proliferation of antigen-specific CD8+ T cells and/or CD4+ helper cells. The binding of PD-L1 to PD-1 or B7.1 transmits an inhibitory signal that reduces the proliferation of antigen-specific T cells in lymph nodes while reducing regulatory T cells (anti-inflammatory, suppressor T cells). This is further mediated by the lower regulation of the gene Bcl-2. The CD274 gene encodes an immunosuppressive receptor ligand expressed by hematopoietic cells and non-hematopoietic cells, and its interaction with its receptor inhibits T cell activation and cytokine production. This interaction is important to prevent autoimmunity by maintaining the homeostasis of the immune response during infection or inflammation of normal tissues. In the tumor microenvironment, this interaction allows tumor cells to immune escape by inactivation of cytotoxic T cells. The expression of this gene in tumor cells is considered as a prognostic factor for many types of human malignancies, including colon cancer and renal cell carcinoma.
