Anti-EGFR Antibody-Cytokine Fusion Protein, TNF-IgG (mAb 425) (CAT#: ACFP-SH080)
This antibody-cytokine fusion protein was achieved by conjugating/fusing the Anti-EGFR IgG to TNFα. It was expressed in CHO and purified with affinity chromatography. The immunocytokine retains the ability to bind the EGFR as well as the biological activity of TNFα. The fusion protein supported mainly proliferation and lymphokine-activated killer cell activity, and TNF-FuP significantly accumulated in the tumor, which led to retardation of tumor growth. This immunocytokine was designed for treating any type of solid or non-solid cancer or malignant lymphoma.

- Tumor necrosis factor (TNF), formerly known as TNFα or TNF alpha, is the best-known member of this class. TNF is a monocyte-derived cytotoxin that has been implicated in tumor regression, septic shock, and cachexia. The protein is synthesized as a prohormone with an unusually long and atypical signal sequence, which is absent from the mature secreted cytokine. A short hydrophobic stretch of amino acids serves to anchor the prohormone in lipid bilayers. Both the mature protein and a partially processed form of the hormone can be secreted after cleavage of the propeptide.
