Anti-MUC1 Antibody-Cytokine Fusion Protein, IgG (Gatipotuzumab)-IL15 (CAT#: VS-0624-FY85)

Gatipotuzumab is a humanized monoclonal antibody that recognizes a tumor-specific epitope of mucin 1 (TA-MUC1), enabling it to distinguish between tumor-MUC1 and non-tumor-MUC1 epitopes. IL-15 intricately governs the activation and proliferation of T and natural killer (NK) cells. Furthermore, IL-15 plays a crucial role in providing the necessary survival signals to sustain memory T cells even in the absence of antigen. The product is a recombinant anti-TA-MUC1-human IL-15 fusion protein, known as GT-00AxIL15. This innovative GT-00AxIL15 immunocytokine consists Fc- and Fab- part of the parental mAb GT-00A, human IL-15, N54Q mutation to remove N-glycosylation site in the Fab domain, Fc N-glycosylation site N297, K447A mutation at transition sequence to IL-15 modules to remove potential protease site; 6xPAPAP linker between IL-15 and the heavy chain.

  • Interleukin 15 (IL-15) is a protein encoded by the IL15 gene. IL-15 is an inflammatory cytokine that is structurally similar to interleukin 2 (IL-2). Like IL-2, IL-15 binds and signals through a complex consisting of the IL-2/IL-15 receptor β chain (CD122) and the common γ chain (γ-C, CD132).

Target

  • Introduction
  • This gene encodes a membrane-bound protein that is a member of the mucin family. Mucins are O-glycosylated proteins that play an essential role in forming protective mucous barriers on epithelial surfaces. These proteins also play a role in intracellular signaling. This protein is expressed on the apical surface of epithelial cells that line the mucosal surfaces of many different tissues including lung, breast stomach and pancreas. This protein is proteolytically cleaved into alpha and beta subunits that form a heterodimeric complex. The N-terminal alpha subunit functions in cell-adhesion and the C-terminal beta subunit is involved in cell signaling. Overexpression, aberrant intracellular localization, and changes in glycosylation of this protein have been associated with carcinomas. This gene is known to contain a highly polymorphic variable number tandem repeats (VNTR) domain. Alternate splicing results in multiple transcript variants.
