This antibody-cytokine fusion protein was achieved by conjugating/fusing the Anti-CD20 scFv-Fc to IL2. It was expressed in CHO and purified with affinity chromatography. The immunocytokine retains the ability to bind the CD20 as well as the biological activity of IL2. This antibody-Fc-hIL2 was at least equivalent to the commercial molecule in both binding to Daudi cells and activation of ADCC and may represent a promising plant-derived biopharmaceutical for the treatment of NHLs. This immunocytokine was designed for treating leukemias, lymphomas, rheumatoid arthritis, and other autoimmune diseases.

In our binding assays with Daudi cells, this molecule performed at least equivalently to commercial anti-CD20 molecules. The scFv-Fc design seems robust and effective for researching treatments for non-Hodgkin's lymphomas.

We observed significant activation of ADCC in our experiments. This antibody-cytokine fusion represents a promising tool for our autoimmune disease research, particularly in modeling rheumatoid arthritis therapies involving B-cell depletion.
