Human Anti-CD59 (clone 1E8) scFv-Fc Chimera (CAT#: VS-0125-FY58)

The recombinant 1E8 scFv-Fc is a single-chain antibody with specificity for CD59. Its design allows for the linkage of both the heavy and light chains of the antibody on the same chain, along with the binding of the human Fc region (CH2-CH3) to enhance its stability and functionality. The recombinant 1E8 scFv-Fc antibody was expressed and secreted into the culture medium after transfection of 293T cells. Flow cytometry analysis revealed that CD59 was highly expressed in prostate cancer cells, and the recombinant 1E8 scFv-Fc exhibited significantly enhanced complement-dependent cytotoxicity (CDC) similar to that of the mouse 1E8 antibody.

Applications

  • Application Notes
  • The anti-CD59 single-chain antibody (scFv-Fc), known as 1E8 scFv-Fc, has the potential to serve as an effective adjuvant therapy in cancer treatment. By targeting CD59-expressing cancer cells, this recombinant antibody may enhance complement-dependent cytotoxicity (CDC), leading to increased tumor cell lysis and death. Given that CD59 is a key regulator of the complement system that inhibits complement-mediated cell death, the binding of 1E8 scFv-Fc to CD59 on the surface of tumor cells could activate the complement cascade. This activation may bolster the anti-tumor immune response, offering a promising avenue for improving cancer therapies.

    Glycated.
Glycation is found in diabetic subjects, but only at minimal levels in nondiabetic subjects.
Glycated CD59 lacks MAC-inhibitory function and confers to vascular complications of diabetes.
    Glycosylation at Thr40, Asn43, Lys66, Thr76, Thr77, Thr105, Ser106, and Thr111
    Modification sites at PhosphoSitePlus
    Glycosylation from GlyConnect CD59_HUMAN (79)
    Glycosylation from GlyGen 7 sites, 216 N-linked glycans (2 sites), 9 O-linked glycans (6 sites)
  • Function
  • Potent inhibitor of the complement membrane attack complex (MAC) action.
Acts by binding to the C8 and/or C9 complements of the assembling MAC, thereby preventing incorporation of the multiple copies of C9 required for complete formation of the osmolytic pore.
This inhibitor appears to be species-specific.
Involved in signal transduction for T-cell activation complexed to a protein tyrosine kinase.
