Provided is an internalizing antibody (clone F4) directed against EGFR (epidermal growth factor receptor), a tyrosine kinase receptor that is overexpressed in a wide range of tumour cells. The clone F4 bound native EGFR-bearing cell lines and could be internalized, but did not bind EGFR-negative cell lines. F4 could be used to target therapeutic agents into tumour cells and was expected to be nonimmunogenic in humans.

F4 p-scFv and s-scFv bound EGFR-positive cell line CHO-EGFR-GFP1, A431, MCF-7 and MDA-MB-231, but did not bind EGFR-negative cell line CHO-K1 and SKOV3.

F4 s-scFv recognized a protein of approx. 170 kDa on A431 cell lysate (lane 2) and CHO-EGFR-GFP1 cell lysate (lane 4), but no band was detected on CHO-K1 cell lysate (lane 3) and prestained protein marker (lane 1).
