Figure 1 AF2 neutralization of IFNγ-mediated induction of MHC class II molecule expression on Hs294T cells.

Figure 1 AF2 neutralization of IFNγ-mediated induction of MHC class II molecule expression on Hs294T cells.

Histograms depict the level of HLA±DR expression (as determined by indirect immunofluorescence using L243) on Hs294T cells that have been incubated in the absence of IFNγ (dotted line), or in the presence of 125 pM IFNγ with (solid line) or without (dashed line) 1 nM AF2.

Thakur, A. B., & Landolfi, N. F. (1999). A potent neutralizing monoclonal antibody can discriminate amongst IFNγ from various primates with greater specificity than can the human IFNγ receptor complex. Molecular immunology, 36(15-16), 1107-1115.

Figure 2 Titration of AF2 neutralization of IFNg activity.

Figure 2 Titration of AF2 neutralization of IFNg activity.

(A) Titration of AF2 neutralization of IFNg activity on Hs294T cells. Graph represents the median fluorescent intensity of HLA±DR expression (as determined by indirect immunofluorescence using L243) on Hs294T cells incubated with IFNg and increasing concentrations of AF2. Dashed line indicates fluorescence intensity of Hs294T cells incubated in the absence of IFNg. (B) Titration of ability of AF2 to inhibit 125I-IFNg binding to Daudi cells. Graph depicts amount of bound 125I-IFNg in the presence of increasing concentrations of AF2. The 125I-IFNg cpm bound in the presence of a great excess (650 nM) of an isotype matched control antibody was 3862±420; 125I-IFNg cpm bound in the presence of 100× (150 nM) concentration of unlabeled IFNg was 510±138.

Figure 4 Neutralization of primate IFNγ by AF2.

Figure 4 Neutralization of primate IFNγ by AF2.

Graph displays the percent of IFNγ neutralization (as indicated by inhibition of HLA-DR expression induction). Hs294T cells were incubated with IFNγ containing supernatant from the indicated primate along with 10 nM AF2. The level of HLA-DR expression in terms of median channel fluorescence in the absence of AF2 was 33 for human, 17 for chimpanzee, 33 for gorilla, 38 for orangutan, 28 for gibbon, 21 for baboon, 31 for rhesus, 15 for marmoset.
